The PERCEPTION pipeline introduces a groundbreaking approach to precision oncology, using single-cell transcriptomics to predict patient responses and resistance to cancer treatments. By integrating large-scale drug screening data with matched bulk and single-cell expression profiles, PERCEPTION builds highly accurate treatment response models tailored to individual tumors. The pipeline has demonstrated success in predicting responses to targeted therapies across cultured cells, patient-derived primary cells, and clinical trials for multiple myeloma and breast cancer. Additionally, it effectively tracks resistance development in lung cancer patients treated with tyrosine kinase inhibitors. Outperforming existing bulk and single-cell-based predictors, PERCEPTION highlights the transformative potential of single-cell omics in personalizing cancer care. Its accessible platform encourages the adoption of single-cell profiling to enhance clinical precision oncology workflows.
